Win32 has been updated for Vista, and continues to be a valid way to program for Windows. 64-bit versions of Windows also support Win64, which is pretty much the same as Win32, except it expects data structures to be 64-bit. It's still the method of choice for doing OS things in C++.
It is, however,
slowly being phased out in favour of more .NET-oriented things. Don't bet on it being truly gone for a long while—give it a few years after Microsoft stops using it internally for people to abandon it. Maybe after Windows 7.0 is finished, they'll get around to deprecating it.
