I don't believe there are any handshaking lines on the programming port. According to the documentation from Parallax and Innovation FIRST, the only pin (other than Tx/Rx/Gnd) connected is DTR which is used to reset the Stamp.

We get an interrupt from our processor when there is data available in the receive buffer.
