What does Stop Build Day mean?

And when exactly is the deadline? Both the manual and the blast leave the date ambiguous. Does it mean Feb 21, 12:00:00am localtime, or does it mean one second after Feb 21, 11:59:59pm localtime?

yes. the bot must go in the bag at the same time the tools must go down.

Yup. At 11:59:59, sealed and ready. But, honestly, no matter how behind we were, we've never had to push it that far. You should be fine, as long as you remember the date.
