Well, actually, since it reads, "This Bond movie had James Bond fighting the title character," (my emphasis) it is, indeed, referencing the title of the movie, not the name of the character.

Thus, "What is Dr. No?" is the appropriate question.

Not that it matters. I hardly ever count grammar against anyone. But it would be "What is?" because I'm asking for the movie.
